We are putting a metal roof on a barn to use for storage and need to be sure it does not leak. We are not sure if we should use caulking in the seams.

This is determined by the type of metal roofing you're installing, and also by the pitch of the roof. Generally, with the type of product I think you're talking about, a tape sealant is a good idea. However, I cannot stress enough -- get full installation instructions from the manufacturer of the roofing. Read and follow them closely! This is critical.
